CRISPR Cas9/gRNA RNP（核糖核蛋白復(fù)合物）由CRISPR RNA(crRNA)和tracrRNA部分雙鏈RNA復(fù)合物及cas蛋白組成。不同于傳統(tǒng)的Cas9/gRNA質(zhì)粒，crRNA和tracrRNA寡聚核苷酸是完整的組分，不需要細(xì)胞自身轉(zhuǎn)錄得到CRISPR相關(guān)組分。直接將Cas9protein 跟sgRNA混合就可以用于轉(zhuǎn)染或者注射。Cas9 RNP與其它系統(tǒng)相比有多個(gè)優(yōu)點(diǎn)：
1）Cas9 RNP在轉(zhuǎn)染后即刻發(fā)生高效率基因編輯
2）Cas9 RNP通過蛋白降解可快速清除以降低脫靶可能性
3）Cas9 RNP是in vivo基因編輯的完美方案
